简介：Fura-2,anaminopolycarboxylicacid,isaratiometricfluorescentdyewhichbindstofreeintracellularcalcium.Itwasthefirstwidelyuseddyeforcalciumimaging,andremainsverypopular.Fura-2isexcitedat340nmand380nmoflight,andtheratiooftheemissionsatthosewavelengthsisdirectlyrelatedtotheamountofintracellularcalcium.Regardlessofthepresenceofcalcium,Fura-2emitsat510nmoflight.Theuseoftheratioautomaticallycancelsoutconfoundingvariables,suchasvariabledyeconcentrationandcellthickness,makingFura-2oneofthemostappreciatedtoolstoquantifycalciumlevels.Thehighphotonyieldoffura-2allowedthefirstrealtime(videorate)measurementsofcalciuminsidelivingcellsin1986.Morerecently,genetically-encodedcalciumindicatorsbasedonspectralvariantsofthegreenfluorescentprotein,suchasCameleons,havesupplementedtheuseofFura-2andothersmallmoleculedyesforcalciumimaging,butFura-2remainsfaster.
