简介：Fura-2-acetoxymethylester,oftenabbreviatedFura-2AM,isamembrane-permeablederivativeoftheratiometriccalciumindicatorFura-2usedinbiochemistrytomeasurecellularcalciumconcentrationsbyfluorescence.Whenaddedtocells,Fura-2AMcrossescellmembranesandonceinsidethecell,theacetoxymethylgroupsareremovedbycellularesterases.Removaloftheacetoxymethylestersregenerates\"Fura-2\",thepentacarboxylatecalciumindicator.MeasurementofCa2+-inducedfluorescenceatboth340nmand380nmallowsforcalculationofcalciumconcentrationsbased340/380ratios.Theuseoftheratioautomaticallycancelsoutcertainvariablessuchaslocaldifferencesinfura-2concentrationorcellthicknessthatwouldotherwiseleadtoartifactswhenattemptingtoimagecalciumconcentrationsincells.
