简介：HEPES(4-(2-hydroxyethyl)-1-piperazineethanesulfonicacid)isazwitterionicorganicchemicalbufferingagent;oneofthetwentyGood'sbuffers.HEPESiswidelyusedincellculture,largelybecauseitisbetteratmaintainingphysiologicalpHdespitechangesincarbondioxideconcentration(producedbycellularrespiration)whencomparedtobicarbonatebuffers,whicharealsocommonlyusedincellculture.Thedissociationofwaterdecreaseswithfallingtemperature,butthedissociationconstants(pK)ofmanyotherbuffersdonotchangemuchwithtemperature.HEPESislikewaterinthatitsdissociationdecreasesasthetemperaturedecreases.ThismakesHEPESamoreeffectivebufferingagentformaintainingenzymestructureandfunctionatlowtemperatures.Lepe-Zunigaetal.reportedaphototoxicityofHEPESwhenexposedtoambientlightbytheproductionofhydrogenperoxide,whichisnotaprobleminbicarbonate-basedcellculturebuffers.ItisthereforestronglyadvisedtokeepHEPES-containingsolutionsindarknessasmuchaspossible.
