简介：Fialuridine,or1-(2-deoxy-2-fluoro-1-D-arabinofuranosyl)-5-iodouracil(FIAU),isanucleosideanaloguethatwasinvestigatedasapotentialtherapyforhepatitisBvirusinfection.Ina1993clinicalstudyattheNIH,unexpectedtoxicityledtothedeathof5outof15patientsfromliverfailureassociatedwithlacticacidosis;twofurtherparticipantsrequiredlivertransplantation.Thistoxicitywasunusualinthatitwasnotpredictedbyanimalstudies.Itissuspectedthatthedrug'stoxicitywasduetoitsdamagingmitochondria.
