简介：Zileuton(tradenameZYFLO)isanorallyactiveinhibitorof5-lipoxygenase,andthusinhibitsleukotrienes(LTB4,LTC4,LTD4,andLTE4)formation.Zileutonisusedforthemaintenancetreatmentofasthma.Zileutonwasintroducedin1996byAbbottLaboratoriesandisnowmarketedintwoformulationsbyCornerstoneTherapeuticsInc.underthebrandnamesZYFLOandZYFLOCR.Theoriginalimmediate-releaseformulationofzileuton,knownasZYFLO,istakenfourtimesperday.Theextended-releaseformulation,ZYFLOCR,istakentwicedaily.
用途：抗白三烯药物.通过对5-脂氧酶的抑制,减少白三烯的生成,用于治疗哮喘.本品为白三烯拮抗剂新药,通过抑制5-脂氧合酶的活性而抑制此酶催化花生烯酸转化成白三烯的过程.已知在诸多炎性介质中,白三烯是引发哮喘的重要因素.所以本品临床用于预防和维持治疗成人及12岁以上少年的慢性哮喘病,另外本品可预防性地减少因阿司匹林敏感的哮喘病人由阿司匹林诱发的支气管痉挛,以及由冷空气和运动引起的气道过度敏感(收缩).
