简介:Endomorphin-2(EM-2)(aminoacidsequenceTyr-Pro-Phe-Phe-NH2)isanendogenousopioidpeptideandoneofthetwoendomorphins.Itisahighaffinity,highlyselectiveagonistoftheμ-opioidreceptor,andalongwithendomorphin-1(EM-1),hasbeenproposedtobetheactualendogenousligandofthisreceptor(thatis,ratherthantheendorphins).LikeEM-1,EM-2producesanalgesiainanimals,butwhereasEM-1ismoreprevalentinthebrain,EM-2ismoreprevalentinthespinalcord.Inaddition,theactionofEM-2differsfromthatofEM-1somewhat,becauseEM-2additionallyinducesthereleaseofdynorphinAand[Met]enkephalininthespinalcordandbrainbyanunknownmechanism,whichinturngoontoactivatetheκ-andδ-opioidreceptors,respectively,andaportionoftheanalgesiceffectsofEM-2isdependentonthisaction.Moreover,whileEM-1producesconditionedplacepreference,ameasureofdrugreward,EM-2producesconditionedplaceaversion,aneffectwhichisdynorphinA-dependent.SimilarlytothecaseofEM-1,thegeneencodingforEM-2hasnotyetbeenidentified.
